The MOAL algorithm applies our previously established DMEA algorithm twice to an input drug screen data frame: first to evaluate if drugs tend to have similar toxicity with other drugs grouped by mechanism of action (MOA) sets and second to evaluate if drugs in each MOA set tend to have similar toxicity to other drugs grouped by MOA sets. MOAL also includes quality control measures. First, you may optionally remove drugs which do not have similar toxicity with other drugs in their own MOA set. Second, MOA pairings only pass our final quality control measures if both MOAs are significantly self-enriched and if both MOAs are significantly enriched for each other in the same direction.
